Sign up whenever, but there's only 9 spots left, so if you want in, better do it soon.
The draft is automatic, and won't occur until at least 4 people have signed up. The sooner the league fills up, the better off we'll be to get our trades done before the season starts.
All drafts (live, auto, offline) must be completed prior to 11:59pm PT on Saturday, September 9 in order to receive scoring for Week 1. Drafts occurring after that time will not be eligible to begin scoring until Week 2.

Now we wait for the rest of the league to fill up so we can get on with the draft/trades. It'll probably be a couple days.
For those who've never played, it's pretty simple, even if you don't know anything about football!
Sign up, there will be a draft, and you will have players on your team. Then you can decide to trade them, waive them, whatever. Come game time (Sept. 10th is 1st game), you'll have a starting lineup. Each player on your team will score points for you based on their performance. Touchdowns, passing yards, sacks, interceptions, etc. are all factors in scoring.
For example, week 1, let's say my team plays keoki's team. If I have crappy players that don't score me points, and keoki's team does well, he'll win that week. It continues much like normal football. At the end (16 weeks!) we'll have playoffs.
So a good practice would be to set up your team for Sunday/monday's game a couple days in advance. There are plenty of online resources that can give FF advice. Who to bench, who to start, etc. These links will be posted on our FF homepage soon.
You do not have to set up your team each week. You just won't do very well if you don't pay attention to it. Players get injured, and injured players won't score the points!

Still one spot left! Once that spot is filled I'll email everyone and let them know (at least a day) in advance when the draft will happen. So someone grab that last spot and let's get this thing going!
